Valerian

Valerian is a natural nootropic ingredient and has been used throughout history as a nootropic to promote feelings of calm, decreases levels of anxiety and stress, and enhances sleep.

Valerian has also been traditionally been used for the treatment of headaches, anxiety, heart palpitations, high blood pressure, irritable bowel, menstrual cramps, epilepsy, and childhood behavior problems.

Valerian helps boost GABA in your brain, inhibits GABA reuptake, modulates serotonin and norepinephrine, and activates adenosine receptors. Valerian acts like an anti-inflammatory by reducing NF-κB (nuclear factor kappa-B). And improves blood flow by acting as a smooth muscle dilator.[i] Valerian helps eliminate stress by boosting GABA, serotonin, activating adenosine receptors, and reducing the stress hormone cortisol.

Melatonin (N-acetyl-5-methoxytryptamine) is known for helping insomnia, is a powerful antioxidant and anti-inflammatory, protects against neurodegenerative diseases, stroke and brain trauma.

It is a hormone primarily produced in the pineal gland. Your pineal gland is filled with cells that respond to light and darkness. Your pineal gland acts as your body’s central clock through its secretion of melatonin. Telling your brain, body and organs when it’s time to be active and when it’s time to rest. This is the reason why melatonin is referred to as the “sleep hormone”.

Serotonin acts as a neurotransmitter on its own. But is also converted into N-acetyl-serotonin by the enzymes serotonin N-acetyl transferase and Acetyl-CoA. Hydroxyindole O-methyl transferase and SAMe convert N-acetyl-serotonin into melatonin through methylation of the hydroxyl group.[vi]

Melatonin prevents neurodegenerative disease. Melatonin improves longevity by increasing the longevity gene called SIRT1. This gene plays an important role in maintaining metabolism and neuroplasticity.[xi]

Melatonin promotes quality sleep. Your pineal gland acts as your body’s central clock by secreting melatonin. This action tells your brain and other organs when it’s time to be active, and when it’s time to rest. Melatonin helps regulate other hormones and maintains your body’s circadian rhythm.[vii]

l-tryptophan

L-Tryptophan is the sole precursor of serotonin, which is often referred to as the “happiness molecule”. L-Tryptophan’s role in brain serotonin synthesis is critical for mood, behavior and cognition. Low serotonin levels can result in pain, insomnia, depression, seasonal affective disorder, and chronic fatigue.

For your body to manufacture serotonin, it needs an adequate supply of the natural amino acid L-Tryptophan. As a nootropic supplement, Tryptophan is used for anxiety, ADHD, depression, insomnia, memory loss, pain, and eating disorders.[i] Tryptophan boosts memory. Tryptophan’s main nootropic mechanism of action is as a precursor to the neurotransmitter serotonin. Other neurotransmitters such as melatonin and beta-endorphin increase following Tryptophan supplementation.[iv] Research shows that Tryptophan and serotonin play a significant role in memory. The cytidine is then converted into uridine, a nucleotide base that is crucial to neural membrane synthesis and is believed to enhance cognition. The released choline acts as a cholinergic, increasing levels of choline in the brain.

Ingredient Specific Contraindications

  • If any surgeries are planned, stop taking valerian at least two weeks before scheduled. Anesthesia and other medications used during surgery also affect the central nervous system like Valerian. The combined effects might be harmful.
  • Citicoline is usually well-tolerated, but side effects may include nausea and diarrhea.
  • Women who are pregnant or breastfeeding, should avoid using citicoline. We do not know if citicoline is safe for these groups.
  • An excess of choline can amplify or trigger depressive episodes, and Citicoline is therefore not advised for people with a predisposition for this.
  • L-tryptophan can interfere with medicines. Check with your doctor if taking any medications.
  • Do not take L-tryptophan if you are on antidepressants known as selective serotonin reuptake inhibitors (SSRIs), MAO inhibitors, tricyclic antidepressants and atypical antidepressants.
  • Women who are pregnant, trying to get pregnant, or breastfeeding, should avoid using Melatonin. We do not know if it is safe for these groups.
  • People with epilepsy, hypertension, bleeding disorders, depression, or transplant recipients should avoid Melatonin.
  • L-Glutamine should not be used by individuals with liver or kidney issues, severe hypertension, MSG sensitivity, seizures, or people with bipolar disorder.
  • Do not take if allergic to gelatin.
